DiO, also called DiOC18(3), is a green fluorescent, lipophilic carbocyanine dye. DiO’s excitation max is 484 nm, and emission max is 501 nm.

DiO is widely used as an anterograde and retrograde neuronal tracer in living and fixed tissues and cells. DiO uniformly labels neurons via diffusion in the plasma membrane. In intact tissue, the dye does not transfer from labeled to unlabeled cells but some transfer may occur when the membrane is disrupted, for example after sectioning.

DiO and DiI are often used together in dual-color studies. But DiO has a slower lateral diffusion rate on the membranes than that of DiI.

DusQ3 is a dark quencher for far red and near infrared fluorophores, especially suitable for Cyanine5.5.

The dye can be attached to 3′-terminus of oligonucleotides by means of automated solid phase synthesis using this support. The dye is sensitive to nucleophilic reagents, and therefore must be carefully deprotected. We recommend the use of ultramild, phenoxyacetyl protected monomers. As an alternative, post-synthetic labeling using NHS ester of the quencher can be used.

DMF (dimethylformamide) is a recommended solvent for labeling reactions using NHS esters. However, when DMF is stored in air, it deteriorates slowly absorbing moisture, and releasing dimethylamine. These impurities rapidly destroy NHS ester function, decrease labeling efficiency, and even make labeling reaction fail.

Dimethylformamide solvent requires special drying and preparation, which is often not available in all laboratories.

This product contains ready-to use solvent which is guaranteed to be useable for the labeling. Tubes with high quality, dry, and amine-free DMF are packed under argon.

AF488 is a fluorescent dye. AF488 is a fluorophore pH-insensitive over a broad pH range (from 4 to 10). It has an absorption maximum at 495 nm and an emission maximum at 519 nm in the green spectrum region. The dye is hydrophilic and can be used to introduce the fluorescent label into various molecules, including proteins and antibodies. Conjugates of molecules with AF488 have high brightness and photostability and are commonly used in flow cytometry and microscopy. This allows the detection of biological objects with high sensitivity at a longer imaging time.

AF488 azide interacts with alkynyl derivatives of biomolecules in Click Chemistry reactions either in the presence of copper (I) catalyst (with terminal alkynes) or without catalyst (with cyclooctynes), leading to the formation of stable adducts.

Pyrene is a polyaromatic hydrocarbon which exhibits fluorescence in blue range of the spectrum. When two pyrene residues are closely spaced, excimer fluorescence is observed at longer wavelength. Therefore, pyrene is useful as a proximity label.

Pyrene exhibits affinity to graphite, and can be used as an anchor group for graphite. This reagent can be conjugated with biomolecules for their subsequent immobilization on graphite surfaces.

This NHS ester allows to label biomolecules with amino groups, such as proteins, peptides, amino-DNA and other molecules.

Pyrene is also a FRET donor for perylene.

Blue emitting Coumarin 343 dye can form a FRET pair with fluorescein (FAM). An amine reactive form, activated NHS ester.

This amine-reactive NHS ester contains an aminohexanoyl linker between the fluorophore, and the reactive group. This linker provides better solubility and spatial separation between the fluorophore, and the target molecule being labeled.

This reagent can be used to design FRET based assays with FAM as acceptor, and to construct systems which harvest blue light energy.
