H2DCFDA (2′,7′-dichlorodihydrofluorescein diacetate) is a common reagent used for investigating the production of reactive oxygen species in living cells.

H2DCFDA is a non-fluorescent fluorescein derivative (its reduced acetylated form). The reagent begins to emit fluorescence only after cleavage of acetyl groups and the reagent’s oxidation in the cell while converting to 2’,7’-dichlorofluorescein. This is a bright green-fluorescent dye (absorption maximum 511 nm, fluorescence maximum 533 nm). This reagent can be used for assays in living cells and is not compatible with sample fixation.

Acetyl groups in H2DCFDA increase its lipophilicity and improve its cell membrane permeability. Once got into the cell, the dye is deacetylated by cell esterases, thus becoming charged and better fixed inside the cell. Oxidation with reactive oxygen forms results in the formation of a fluorescent product (2’,7’-dichlorofluorescein) and can be detected using various methods, for example with a flow cytometer, plate reader, or fluorescent microscope.

Recommendations for using the reagent使用试剂的建议::

  • Use a freshly prepared solution of the reagent (the working solution is not intended for long-term storage because of gradual reagent oxidation).
  • Select an optimal working concentration of the reagent and incubation time required for reagent deacetylation and oxidation for the specific cell line and assay conditions. If there are no protocols recommended for the specific cell line, start with a concentration from 1 to 10 µM and incubation for 30 min.
  • Do not incubate the dye with the cells in the presence of serum because it contains enzymes that cleave H2DCFDA.

Controlled pore glass solid supports are used for the synthesis of 3′-labeled oligonucleotides. This solid support is intended for the synthesis of oligonucleotides bearing fluorescein (FAM) fluorescent dye on 3′-terminus of oligonucleotide. It contains pure 6-isomer of fluorescein. The structure of the reagent is based on chiral, enantiomerically pure scaffold of hydroxyprolinol.

The solid support ensures optimal yield of oligonucleotides up to 60mer. For longer oligos, CPG 1000 should be used. The reagent is compatible with standard ammonia cleavage and deblocking conditions.

This phosphoramidite allows fluorescein (FAM) to be introduced into any position in the oligonucleotide sequence during synthesis by the phosphoramidite method (in the middle of the chain, at the 5’- and 3’-terminus). The reagent is a conjugate of deoxythymidine phosphoramidite and 6-isomer of FAM. Modification is performed during oligonucleotide synthesis by substituting standard dT phosphoramidite with fluorescein-dT phosphoramidite. This modification does not affect exonuclease or polymerase activity.

For modification with fluorescein at the 5’-terminus use FAM phosphoramidite, 6-isomer.

Usage

Coupling: 10 minutes

Deprotection: standard conditions using ammonium hydroxide; deprotection time depends on oligonucleotide composition and nucleobase protecting groups (deprotection for 17 h at 55 °С removes all protecting groups from standard nucleobases). AMA (solution of 30% ammonium hydroxide/40% aqueous methylamine 1:1 v/v) can be used with ~5% of non-fluorescent side product forming. To avoid formation of the side product, start deprotection with ammonium hydroxide (30 min at room temperature), then add an equal volume of 40% aqueous methylamine and continue deprotection as required with AMA (e.g. 10 min at 65 °C).

6-FAM-11-2’-deoxyuridine-5’-triphosphate, trilithium salt, is a common agent for non-radioactive DNA labeling.

FAM (fluorescein) is a popular green-fluorescent dye with an emission maximum at 513 nm. This derivative is a pure FAM 6-isomer.

6-FAM-11-dUTP is a modified triphosphate that can be incorporated into DNA instead of dTTP by various enzymes, including Klenow fragment, Taq-Polymerase, DNA-Polymerase I, Phi 29 Polymerase, reverse transcriptase or terminal transferase.

In contrast to triphosphates with a dye directly linked to aminoallyl-dUTP, this triphosphate contains a linker of 11 atoms between the fluorophore and the nitrogenous base. This linker length prevents FAM-label from potential static quenching and increases the efficiency of nucleotide incorporation during DNA synthesis.

6-FAM-11-dUTP can be used to produce a labeled product during PCR and cDNA synthesis and for labeled DNA-probes synthesis by random primed labeling and Nick-translation. Synthesized fluorescence-labeled DNA probes can be used to detect specific sequences by Southern blot, Northern blot, in situ hybridization, or by microarray analysis.

6-fluorescein (FAM) derivative of deoxycytidine triphosphate (dCTP).

FAM 6-isomer is a common fluorophore used for labeling biomolecules; FAM has a high quantum yield and bright green emission.

FAM-11-dCTP can be used for enzymatic DNA labeling via Nick-translation, 3’-end labeling, random primed labeling, PCR, or cDNA synthesis. FAM-11-dCTP is incorporated into the growing DNA chain by Taq-polymerase, DNA-polymerase I, Phi 29 polymerase, Klenow fragment, reverse transcriptase, or terminal transferase.

The linker length (11 carbon atoms) is optimal for the efficient incorporation of fluorescently-labeled dCTP into the growing DNA chain.

Labeled DNA can be used for FISH experiments, microarray gene profiling, Southern blot, and Northern blot.

Fluorescein (FAM) is one of the oldest yet well known fluorescent dyes. This derivative of FAM contains cyclooctyne group (dibenzocyclooctyne, abbreviated as DBCO or ADIBO) for the conjugation with azides via copper-free, strain-promoted azide alkyne cycloaddition (spAAc).

FAM DBCO can be used for the labeling of proteins, peptides, nucleic acids, and other molecules containing azide groups. Azides react with DBCO spontaneously upon simple mixing. The reaction does not require any catalysts, and it is very efficient.

Fluorescein can be detected using a variety of different instruments.
