Dibenzocyclooctyne sulfotetrafluorophenyl ester (DBCO STP ester) is a water-soluble reagent that can be used for the modification of peptides, antibodies, proteins, and other molecules containing the -NH2 group.

STP esters are hydrolyzed much more slowly than NHS esters, thus allowing efficient biomolecule modification in the aqueous medium.

The dibenzocyclooctyne (DBCO) moiety, which is also known as ADIBO or DIBAC, is the most common substrate for copper-free Click Chemistry reactions that are promoted by ring strain. In this reaction in the presence of a Cu(I) catalyst, DBCO derivatives react with biomolecules functionalized by the azide group to result in stable triazoles.

Dibenzocyclooctyne (ADIBO, DBCO) is one of the most reactive cycloalkynes for strain promoted alkyne azide cycloaddition (spAAC) – a copper free Click chemistry reaction.

This is an amine-reactive NHS ester that provides easy attachment of the reactive moiety to almost any primary or secondary amine group, such as that of protein, peptide, or small molecule amine.

DBCO reacts instantly with azides. The rate of the reaction is much higher than that of copper-catalyzed reaction, and reactions with many other cyclooctynes. Unlike some other cyclooctynes, DBCO does not react with tetrazines – this allows to carry out orthogonal conjugation of azides with DBCO, and trans-cyclooctenes with tetrazines.

Sulfo-Cyanine5.5 is a dye with far-red emission approaching NIR range. It has found application in non-invasive live-organism imaging. This particular derivative of sulfo-Cyanine5.5, the DBCO (or ADIBO) derivative, contains cycloalkyne for copper-free conjugation of this fluorophore with various organic azides.

The reaction between DBCO and azides is blazingly fast, orders of magnitude exceeding the rate of copper-catalyzed reaction between azides and terminal alkynes. It is also catalyst-free.

This reagent can be used to conjugate sulfo-Cyanine5.5 dye to a variety of azide-labeled molecules.

Strained cycloalkynes, such as cyclooctynes, react with azides very rapidly in the absence of copper catalyst in a strain promoted alkyne azide cycloaddition (spAAC). This reaction is very fast, mild, and biocompatible.

Compared to other cycloalkynes, DBCO provide among the fastest reaction kinetics, still possessing good stability.

Cyanine3.5 is a bright and photostable fluorophore, which is similar in its spectral characteristics to Cy3.5™ dye. Cyanine3.5 DBCO fluorescence maximum is at 604 nm and in the orange-red spectrum range.

Cyanine3.5 fluorescence is pH insensitive in the range from 4 to 10 and can be detected in the spectrum range with a low level of autofluorescence of biological samples.

Cyanine3.5 DBCO (dibenzocyclooctyne) easily forms conjugates with azide derivatives of biomolecules in copper-free Click Chemistry. The process runs without catalysts or increased temperature, resulting in stable triazoles.

Conjugates of biomolecules with Cyanine3.5 can be used for various microscopy assays, including FRET-microscopy, and in proteomics.

BDP TR is a bright and photostable red-fluorescent dye. Because of its hydrophobic properties, BDP TR can readily permeate cell membranes and is an excellent fluorophore for staining various lipophilic compounds. This dye is an analog of BODIPY™ TR and similar to  ROX by its spectral characteristics.

BDP TR has a long excitation lifetime; besides conventional fluorescence microscopy, it can be used in various fluorescence anisotropy assays.
