Iodo-PEG3-Azide contains a highly reactive iodide functional group which can alkylate a variety of C-, O-, N-, S-centered and other nucleophiles.

Organic iodides are roughly an order of magnitude more active than bromides, and four orders of magnitude more active than chlorides in SN2 reactions. This means that the same alkylation will take 10x less time with iodide than with bromide. Therefore, this molecule helps to effect very fast alkylation.

PEG3 is a moderately long hydrophilic linker. The azide group participates in Click chemistry reactions. It is also easily reduced to an amine group.

Biotin azide is a derivative of biotin (vitamin H) covalently bound with azide group. This reagent allows labeling of various alkynylated molecules (such as DNA, oligonucleotides, and proteins) with biotin. Biotin binding to avidin or streptavidin can be used in downstream affinity applications for the isolation of biotinylated molecules or their binding with streptavidin conjugates.

Azidomethyl-dU (AmdU) is a nucleoside that contains an azide group. The structure of the nucleoside is similar to thymidine, and it is incorporated into nascent DNA by cellular polymerases, similarly to EdU.

Unlike EdU that needs to be further modified with azides in the presence of copper catalyst, AmdU can also react in the absence of copper catalyst using spAAC Click Chemistry with strained cycloalkynes, such as cyclooctynes. This enables detection of nascent DNA in benign, copper free conditions.

Tetramethylrhodamine (TAMRA) is a xanthene dye with orange emission. The dye is a FRET acceptor for FAM, and sometimes used as a quencher for it.

Like other xanthenes, TAMRA exists as two isomers (5- and 6-), which have very similar spectral properties. This is an azide derivative of 6-isomer of TAMRA. The azide can be conjugated with terminal alkynes using copper-catalyzed Click chemistry, or with cycloalkynes with copper-free strain promoted alkyne azide cycloaddition (spAAc) reaction.

Sulfo-Cyanine5.5 is a water-soluble, hydrophilic fluorophore which possesses emission in far red area of the spectrum, an analog of Cy5.5®. As other cyanine dyes, sulfo-Cyanine5.5 has an outstanding molar extinction coefficient which gives rise to its bright fluorescence.

The molecule contains four sulfo groups that provide hydrophilicity and negative charge to the fluorophore – this minimizes non-specific binding.

The azide group of sulfo-Cyanine5.5 azide can be conjugated with terminal alkynes in the presence of copper(I) catalyst, or with cycloalkynes in copper free strain promoted reaction.

Sulfo-Cyanine3.5 is a fluorescent dye with absorption and emission spectra between those of Cyanine3 and Cyanine5. Because of its spectral characteristics, this fluorophore is widely used in BRET and FRET experiments both as a pair donor and acceptor.

Sulfo-Cyanine3.5 is a sulfonated derivative of Cyanine3.5 dye because it contains four additional sulfo groups, which ensures high water solubility of the dye and its conjugates. This allows working with this reagent in aqueous solutions without adding organic solvents.

This reagent is an azide derivative of sulfo-Cyanine3.5 dye for Click Chemistry reactions. The azide group allows conjugating the fluorophore with molecules containing alkyne or cycloalkyne groups, including various biomolecules, small molecules, and polymers, using copper-catalyzed Click chemistry and copper-free spAAC reaction.

Pyrene is a polyaromatic hydrocarbon with strong short-wavelength fluorescence. Unlike other fluorescent dyes, polyaromatic hydrocarbons are fluorescent probes with strong sensitivity to microenvironment. Thus, its fluorescence is different in polar, and nonpolar environments. Other effects can also be observed.

When two pyrenes are in close proximity, they form excimers. Excimer formation can be easily observed, and quantitatively estimated using fluorescent spectra.

Pyrene azide is a reagent for easy pyrene Click Chemistry labeling of any alkyne-bearing molecule. It allows to turn any molecule into pyrene-bearing probe.

This azide contains hydrophilic triethyleneglycol linker to mitigate intrinsic pyrene hydrophobicity, and facilitate attachment to biomolecules in aqueous solutions.

Hexachlorofluorescein (HEX) is a hexachlorinated derivate of the fluorescent dye fluorescein. HEX-labeled oligonucleotides are used in PCR and the HEX channel is widely used in multiplex qPCR.

Oligonucleotides with a HEX label can be easily generated using HEX azide and alkyne-containing oligonucleotide via azide-alkyne cycloaddition.

HEX exhibits an excitation peak at 533 nm and an emission peak at 549 in green-yellow area, and it can serve as a substitute for JOE, BODIPY™ 530/550, VIC™ because of the similar spectral characteristics.

Cyanine7.5 is a NIR dye with long-wave infrared fluorescence. This derivative is azide for Click Chemistry.

Can be used for the construction of various labeled biomolecules containing Cyanine7.5, near infrared fluorescent dye, and an improved analog of Cy7.5®. These conjugates can take advantage of NIR tissue transparency when used for in vivo imaging. This fluorophore is also useful for other fluorescent applications, especially requiring low fluorescent background.

Azide is available as DMSO solution, ready for general Click Chemistry labeling protocol, or in solid form for custom labeling applications.

Cyanine5 azide labeling reagent for Click Chemistry, available as 10 mM solution in DMSO, and in solid form.

This azide is soluble in organic solvents (e.g. DMSO, DMF), therefore the labeling reaction should be carried out with a small amount of an organic co-solvent. This azide can be used for the labeling of alkyne-modified biomolecules in mixtures of water with organic solvents. The solution in DMSO is ready for use in bioconjugation. A water-soluble sulfonated version of this reagent is also available.

Cyanine5 is an analog of Cy5®, one of the most commonly used fluorophores which is compatible with various instruments. Cyanine5 can also be used as a replacement for DyLight® 649.

Cyanine3 dye azide for Click Chemistry, an analog of Cy3® azide. Cy3® is one of the most broadly used fluorophores which can be detected by various fluorometers, imagers, and microscopes. Due to inherently high extinction coefficient, Cyanine3 is also easily detected by naked eye on gels, and in solution. This is non-sulfonated dye which requires organic co-solvent (DMF, DMSO, or other) for efficient labeling in water. Water-soluble version of this reagent is also available.

Product is available both as solid compound, and as 10 mM solution in DMSO which is ready to use in our recommended protocol.

Cyanine3 fluorescent properties are identical to Cy3®, and similar to DyLight 549.

Coumarin 343 is a blue emitting fluorophore with an emission maximum around 480 nm. This dye forms a FRET pair with fluorescein, and can harvest blue light energy for the subsequent transfer to other fluorophores.

The azide derivative can be conjugated with alkynes in a copper-catalyzed and copper-free Click chemistry reactions. The molecule contains a long aminohexanoyl linker that provides separation between the dye, and the azide function.

This is an azide for copper catalyzed and copper free Click chemistry labeling with BDP TR, a bright red emitting fluorophore. BDP TR is an alternative to ROX. This fluorophore is based on borondipyrromethene scaffold, and contains aryl substituents to match its absorption and emission to ROX channel.

ROX dye is prone to oxidation, BDP TR is much more resilient to oxidants. Its brightness and photostability is also very high.
