概述
Exo-CIP 快速 PCR 纯化试剂盒含有热敏的核酸外切酶 I（Exo I）和热敏小牛肠磷酸酶（CIP），用于 PCR 扩增后快速去除残留的 PCR 引物，并对冗余的 dNTPs去磷酸化。PCR产物中存在过量的引物或 dNTPs 可能会影响后续分析，例如会妨碍用紫外光谱法（如 Nanodrop™）对dsDNA 的准确定量。反应在 37°C 下仅需 4 分钟，然后在 80°C 下加热 1 分钟即可对酶迅速灭活。整个反应过程仅需 5 分钟，PCR 产物就可用于下游实验，如 Sanger 测序、SNP 检测或 NGS 文库制备。Exo-CIP 快速 PCR 纯化试剂盒适用于几乎所有 PCR 反应体系。

概述
T4 多聚核苷酸激酶能够催化 ATP 的 γ-位磷酸基团转移到寡核苷酸链（双链或单链 DNA 或 RNA）的 5´ -羟基末端以及 3´ -单磷酸核苷上。T4 多聚核苷酸激酶（NEB #M0201）还具有 3´ 磷酸酶活性，将 3´ -磷酸基团从寡核苷酸的 3´ 磷酸末端、脱氧 3´ -单磷酸核苷和脱氧 3´ -二磷酸核苷上水解掉。经修饰后的酶（NEB #M0236）具有所有激酶的活性，但是缺失了 3´ 磷酸酶活性。
来源
重组 E. coli 菌株，克隆有 T4 多聚核苷酸激酶或修饰的 T4 多聚核苷酸激酶基因。
反应条件
1X T4 多聚核苷酸激酶反应缓冲液
[70 mM Tris-HCl（pH 7.6 @ 25℃），10 mM MgCl2，5 mM DTT]，37℃ 温育。
热失活：65℃ 加热 20 分钟。
注意事项
达到最佳酶活力，需要新鲜缓冲液（在旧缓冲液中，由于氧化造成的 DTT 含量减少会降低酶活力）。
放射性标记反应
50 μl 反应体系中，用 1X T4 多聚核苷酸激酶反应缓冲液、50 pmol 的 γ-[32P] ATP 和 20 单位的酶 37℃ 温育 30 分钟可催化 1-50 pmol 的 5´ 末端磷酸化。[33P] ATP 可代替 [32P] ATP 作标记反应。
CTP、GTP、TTP、UTP、dATP 及 dTTP 均可替代 ATP 作为磷酸供体。

要提高平齐末端或 5´ 凹陷末端的磷酸化效率，可在加入 T4 多聚核苷酸激酶前，先将 DNA 溶液于 70℃ 加热 5 分钟，然后冰上冷却，并加入 5%（W/V）的 PEG-8,000。
T4 多聚核苷酸激酶需要 ATP 才能发挥活性，但是为了适用于高活力的放射性标记反应，在随酶提供的反应缓冲液中不含 ATP。
一般来说，进行激酶反应之后是连接反应。在这种情况下，T4 多聚核苷酸激酶反应在连接酶反应缓冲液中 37℃ 温育 30 分钟即可。反应后的产物可直接进行连接，无需改变缓冲液和进行热失活。如果连接时需要保持其它 DNA 片段的去磷酸化状态，则需要在连接反应前热失活 T4 多聚核苷酸激酶。
在 50 μl T4 多聚核苷酸激酶反应缓冲液体系中，37℃温育 30 分钟，γ-磷酸基团从 ATP 转移至 d(T)8 的 5´ 羟基末端 [ATP 与 d(T)8 比例是 1:1，量为 50 pmol]，结果显示 20 单位酶量可使超过 90% 的磷酸基团转移。

概述
小牛肠碱性磷酸酶（CIP）催化 DNA 和 RNA 5´ 和 3´ 磷酸单脂的去磷酸化反应。另外，CIP 水解核糖和脱氧核糖核苷三磷酸（NTP 和 dNTP）。CIP 在分子生物学研究中应用广泛，如去除 DNA 和 RNA 末端的磷酸基团，用于后续的克隆和探针末端标记。在克隆中，对线性载体去磷酸化，防止自连。该酶可以作用于 5´ 突出端、凹陷端和平末端。CIP 也可以用来降解 PCR 反应中的游离 dNTP，用于制备测序模板和 SNP 分析。

概述
NEBNext 接头是为 DNA、ChIP DNA 和 RNA（不包括 Small RNA）文库构建而设计的，能够确保接头的高效连接及文库的高产量，并且最大限度的减少接头二聚体的形成。NEBNext 接头包含一个特殊的发卡环状结构，能够更高效的和经末端修复的带 dA 尾的 DNA 结合。环状结构包含一个 U，当 U 被 USER 酶（由 UDG 和内切酶 VIII 组合而成）切掉后，环状结构打开，使它可以成为 PCR 的反应底物。检索序列通过 PCR 引入文库，从而实现了多样本的制备。NEBNext 接头引物不仅能够用于 NEBNext 产品，也可以用于其它的兼容 Illumina 标准平台的文库制备方法。
提供单端和双端检索引物。同时也提供用于重亚硫酸盐测序的甲基化接头。
功能验证
每个试剂盒都通过对基因组 DNA、ChIP DNA 和 mRNA 进行文库构建，并在 Illumina 平台测序进行验证。
