1、使用方便:

厌氧培养

在厌氧罐或可立培养袋中使用

对于培养量比较大的情况,使用anaerobic jars厌氧罐或者W-Zip standing Pouch自封可立袋。选择合适体积的厌氧产气袋即可。

-厌氧产气袋可以快速提升二氧化碳浓度,达到厌氧环境,有利于得到较大的菌落;
-特别对于布鲁氏菌,卟啉单胞菌的培养,Anaero Pack产气袋是被文献报道过的最佳培养方法;
-由于产气袋不会造成容器内压力有大的变化,以及不会提高温度,产气袋可以放在通用的厌氧罐内,打开和关闭厌氧罐非常简单;
-产气袋的主要成分是抗坏血酸(维生素C)。

 使用方法  将产气袋的铝塑包装上侧剪开,取出纸袋,纸袋不需要打开,立即放入培养容器内,马上密封容器
 注意事项  ①当产气袋拆开包装后,就立刻和周围的氧气进行反应,应马上放入厌氧罐或培养袋中,然后立即
盖上厌氧罐盖子(1分钟内)或把培养袋密封(30秒内);
②当在同个容器内使用2个及以上产气袋时,不要把它们堆叠在一起;
③使用过的产气袋有时候会因为剩余反应而产生热量,应让它们恢复到室温30分钟后再丢弃;
④对于嗜热菌,应将培养容器放入适当温度的培养箱内,并选择占用的产气袋A-05。

微需氧培养

使用Anearo Pack 微需氧产气袋后的气体浓度变化

在1小时内,氧气浓度变为6-12%,二氧化碳浓度变为5-8%,形成微好氧环境

氧气:维持在6-12%适合微好氧微生物的培养
微需氧细菌比如弯曲杆菌,螺旋杆菌在氧气浓度大于15%的条件下很容易死亡,当氧气浓度小于5%,菌落变小,微需氧产气袋可以吸收容器内的氧气,在1小时内使氧气浓度为6-12%,然后微好氧微生物可以自然生长。

二氧化碳:低于10%,不影响pH
适合微需氧细菌培养的二氧化碳浓度为不低于5%,更高一点的二氧化碳浓度并不会太影响培养结果,相反,溶解的二氧化碳会降低培养基的pH值,对于一些抗生素敏感测试会产生不利的影响。Anearo Pack微需氧产气袋会通过将二氧化碳浓度维持在5-8%来阻止二氧化碳浓度过高的情况。

对于小型培养袋和小型厌氧罐

使用350ml微需氧产气袋/小型微需氧产气袋,可以培养2个皿。

当只需要培养1个皿时,使用一个空皿一起放进去,以控制气体大小,达到最佳产气效果。

注意:
弯曲杆菌和螺旋杆菌喜欢较高的湿度;
比如可以蘸取20ml水在棉絮或者滤纸上,放进厌氧罐内以避免琼脂表面过于干燥;
注意不要碰到产气袋,采用有间隔的厌氧罐是比较适合的。

二氧化碳产气袋造成约5%二氧化碳浓度,适合嗜二氧化碳细菌的生产,比如嗜血杆菌属和奈瑟氏菌属等。

可用于研究致病菌的RTI指数,比如链球菌和黏膜炎奈瑟氏球菌,据报道,肺炎链球菌和米勒链球菌在二氧化碳产气袋下培养比用蜡烛箱效果好。

特殊培养条件的产气袋

三菱MGC同时有特殊应用的产气袋:

当进行超过45℃的高温厌氧细菌培养时,普通的厌氧产气袋不适合使用,可以使用专用的高温菌用厌氧产气袋。

当进行细胞培养实验时,也可以选择三菱的厌氧5%产气袋,可通过低成本的耗材,即可实现低氧细胞培养。

当培养的细菌不需要过多的二氧化碳时,也可以选择厌氧10%产气袋和厌氧5%产气袋进行培养。

使用方法

标准型,薄型,大型厌氧罐都有间隔仓:

A:放置产气袋;B:放置厌氧指示剂,也可放蘸20ml水的湿棉球/滤纸

盖上盖子时,要两侧同时扣紧,不能先扣一边再扣另外一边,以免损坏盖子

罐体材料:Polycarbonate PC材质,外观透明,可耐受温度:-50℃~140℃

密封条材料:Polyacetal 可耐受温度:-30℃~140℃

注意:抗热性和耐冷性是材料本身的性质,压力的变化可能会损坏厌氧罐;
不能高压灭菌;
培养后可能遇到罐内压力变化无法打开盖子,这时候要多用一些力打开;
培养嗜热菌时,不能使用厌氧罐,要使用立式培养袋;
厌氧罐脏了之后可以用洗涤剂或者酒精洗涤。

自立袋 2.5L可立培养袋

自封袋可最多放12个90mm培养皿,在使用袋夹的情况下,最多可以放置14个90mm培养皿

建议使用培养皿支架进行微生物培养。

注意事项:

虽然产气袋是一次性的,但是培养袋可以重复使用,除非出现破损;

厌氧指示剂(氧气指示剂)

虽然厌氧指示剂的颜色变化是可逆的,但是其灵敏度会下降,因此建议一次性使用;

厌氧指示剂的包装膜上有针孔,氧气会进入针孔导致颜色变化,按照原样使用,不要将药丸取出;

厌氧指示剂应冷藏2-8℃,保持在包装袋内密封,当使用其中1个时,另外的部分应立即密封;

在适当的冷藏条件下,可以保存6个月。

GasPakTM EZ Pouch 孵育袋是一完整的产气系统,可产生高二氧化碳的厌氧环境,适用于初步分离、培养厌氧菌。减少水蒸气产生,透视度极佳。所有产生厌氧环境所需物品均含在内,包括指示剂。仅需一个加入产气袋的步骤,就可启动反应。

内附的活化剂会释放出柠檬酸(Citric acid),产生酸性环境,同时与无机碳酸盐反应而产生二氧化碳。再者,它也会启动铁粉的氧化反应,形成铁锈,快速的将氧浓度降低至小于1%。

在整个反应过程中不会产热,也不需使用催化剂。

每一个厌氧孵育袋可放1-4个培养皿,每一个微需氧和CO2孵育袋可放2个培养皿。

GasPakTM Pouch的密封棒可提供有效的密封效果,不必另外使用电热式密封机。

Merck的厌氧产气系统,包括了2.5L厌氧罐,2.5L厌氧产气袋,厌氧指示条等产品。

2.5L厌氧罐 Anearojar

2.5L体积,可容纳 12 个一次性塑料培养皿。罐子的聚碳酸酯基座通过四个弹簧夹固定在盖子上。在不太可能发生压力的情况下,夹子本身允许自动通风。还提供了携带把手,使得罐子能够在工作台和培养箱之间安全地运输。

易于操作的真空释放螺钉允许空气进入罐中,从而克服了由于存在负压而可能引起的任何困难。

Anaerojar 重量轻、坚固耐用,具有出色的节省空间的特点;没有外部夹子或突出的活门,把手塞进罐子本身的盖子里。

Anaerogen 是与 Anaerojar 一起使用的套件。它在不使用水或催化剂的情况下快速产生厌氧气。不产生氢气。氧气被迅速吸收,而不会产生压力。最终的气体中含有少于 1% 的氧气,补充了二氧化碳——非常适合厌氧菌的最佳生长和增加的菌落大小。

厌氧产气袋是一种高度创新的产品,专用于安全、快速地为厌氧生物创造最佳生长条件。无需加水和催化剂,不产生氢气。厌氧产气在30分钟内完成。

含有亚甲基蓝染料,在厌氧环境下变色无色,氧气恢复后变回为蓝色,用于指示厌氧罐环境中是否纯在氧气。

Description

The CrystalCap SPINE HT is a magnetic sample mount (also known as a cap, pin or goniometer base) designed for cryocrystallography systems that accept SPINE style caps. The CrystalCap SPINE HT attaches to a magnetic CrystalCap SPINE Vial and magnetic goniometer base. The tip of the CrystalCap SPINE HT accepts a Mounted CryoLoop™.

The CrystalCap SPINE Vial is a 1.8 ml cryo vial featuring two small vents and is compatible with the CrystalCap SPINE HT. A ring magnet is molded into the open end of the vial so that when the cap is positioned in the vial, the ring magnet holds the cap on the vial during cryogenic storage. The CrystalCap SPINE Vial features chamfered edges for enhanced cap positioning as well as a magnetic alloy bottom for stability. The HR8-114 CrystalCap SPINE Vial does have a magnet on the bottom of the vial.

The hollowed out design of the CrystalCap SPINE HT is compatible with SPINE style grippers, auto mounters and sample handlers.

The CrystalCap SPINE HT features a two dimensional (2D) alphanumeric 16 x 16 data matrix code on the underside of the cap. Each cap is also color coded for CryoLoop size.

CrystalCap SPINE without 2D code/printing (HR8-094) is the CrystalCap SPINE HT cap only, without 2D code, without color code, without white white background on bottom of cap and no alphanumeric side labeling; cap only, without Mounted CryoLoop.

Note: Caps with Mounted CryoLoops are sold without vials. Vials available separately.

Description

The Seed Bead kits are used to create a seed stock for performing subsequent seeding experiments. The Seed Bead kits contains 24 Seed Bead Tubes, a special 1.5 ml microcentrifuge tube, shaped to maintain the seed in the sample during mixing. Each tube is configured with a PTFE, Stainless Steel, Zirconium Silicate Ceramic, or Glass Seed Beads.

Seeding allows one to grow crystals in the metastable zone. Crystallization in this zone provides control, reproducibility and an improved likelihood of a successful crystallization experiment. Also, crystals can grow from seeds but cannot spontaneously nucleate. By placing a seed or solution of seeds in a drop which is saturated to the metastable zone, one can use the seeds to grow larger single crystals. By controlling the number of seeds introduced into the drop, one can control the number of crystals grown. It is not practically possible to measure and know the number of seeds introduced to a drop, but by performing serial dilutions from a concentrated seed stock, one can control the number of crystals grown in the drop.

Using the Seed Bead kits, one can create crystal seed stock for subsequent seeding experiments. Crystals are placed in the microcentrifuge tube with the Seed Bead(s) and either vortexed or sonicated to generate a seed stock. Then by performing serial dilutions, one can control the number and size of crystals in the experiment.

The Seed Bead kit is useful for the preparation of seed stocks for automated and semi-automated microseeding (D’Arcy 2007, Harlos 2008).

Each Seed Bead Kit (HR2-320) contains 24 special microcentrifuge tubes, each tube containing a single 3mm PTFE Seed Bead.

Each Seed Bead Steel Kit (HR4-780) contains 24 special microcentrifuge tubes, each tube containing a six 1.6 mm Stainless Steel Seed Beads.

Each Seed Bead Ceramic Kit (HR4-781) contains 24 special microcentrifuge tubes, each tube containing a ten 1.0 mm Zirconium Silicate Ceramic Seed Beads.

Each Seed Bead Glass Kit (HR4-782) contains 24 special microcentrifuge tubes, each tube containing a ten 1.0 mm Glass Seed Beads.

These optically clear tapes are compatible with protein crystallization reagents. The polypropylene Crystal Clear Sealing Tapes use acrylic solvent based adhesives which are compatible with aqueous crystallization reagents.

Catalog number HR3-511 is a 1.88 inch (48 mm) wide, 3 mil tape on a 43.7 yard (40 M) roll with a 1.5 inch core and is supplied with a green dispenser / cutter.

Catalog number HR4-511 is a 1.88 inch wide, 3 mil tape on a 109.3 yard roll with a 3 inch core and no dispenser / cutter.

Two strips of the HR3-511 or HR4-511 will seal a Corning, Cryschem, CrystalClear Strip, Greiner, Intelliplate, Linbro, Swissci (MRC), VDX and VDXm plate.

Description

The LCP Sandwich Set consists of a base glass slide and an optimized cover slip. LCP Sandwich Set developed jointly with the renowned Scripps Research Institute in La Jolla, California, USA and is manufactured by Paul Marienfeld (0890003).

The LCP Glass Sandwich Set is a specially designed plate for either automated or manual setting of 96 Lipidic Cubic Phase matrix screening experiments. The plate can also be used for the bicelle method and batch method. The thin (< 2 mm high) plates have exquisite optical properties and are well suited for the detection of microcrystals and for birefringence-free imaging between crossed polarizers. The plates allow for in meso crystallization trials with 50 nanoliters protein/lipid mesophase and 1 microliter precipitant solution per trial.

The 96 well LCP Glass Sandwich Plate consists of a) 127.8 x 85.5 x 1 mm glass base plate with the footprint of an SBS-compliant plate, a 140 µm thick double sticky spacer with 96 punched out holes and b) a 0.2 mm thick glass coverslip. The double sticky spacer is already adhered to the base plate. A brown paper liner covers and protects the top of the double sticky spacer and base plate. The 0.2 mm thick glass coverslip fits over and seals the entire 96 well lower plate. Alternatively a series of twenty-four siliconized 18 x 18 mm No. 1 square cover slides (HR3-152) can be used to seal the entire 96 well lower plate. Each 18 x 18 mm No. 1 cover slide seals 4 wells. Space for a bar code is available at the left end of the plate. Each well can contain 50 nanoliters of cubic phase and 1 microliter of crystallization reagent.

18 x 18 mm No. 1 Siliconized Cover Slides are made from chemically resistant borosilicate glass D 263 M of the first hydrolytic class. The slides are colorless, clear, and suitable for fluorescence microscopy. The slides feature a super hydrophobic surface on both sides. The slides are No. thickness (0.13 ro 0.16 mm). The slides are supplied in a two compartment plastic box, 100 slides per box, 10 boxes per case, 1,000 slides total for HR3-152.

The Tungsten-carbide glass cutter can be used for cutting and removal of the upper coverglass that seals the LCP Sandwich Set. Sold separately.

Description

FRAP (Fluorescence recovery after photobleaching) measures membrane protein mobility in lipids in a high throughput fashion. The system utilizes an automated high throughput LCP-FRAP assay to prescreen membrane protein crystallization conditions.

The FRAP assay can be used to pre-screen multiple protein constructs, lipids and additives to eliminate those conditions that are non-conducive to protein diffusion, and thus crystallization, from the subsequent crystallization trials. High protein mobility and fast diffusion rates correlate well with crystallization conditions.

The FRAP sandwich set consists of a base glass slide with sealing film and an optimized cover slip. This product has been developed jointly with the renowned Scripps Research Institute in La Jolla, California, USA.

The FRAP Sandwich Set is optimized for the pre-crystallization study of membrane proteins.

Developed at the NIH Roadmap Center for membrane protein research and routinely used in GPCR structural biology.

Bottom Slide Features
• Dimensions: approx 127.8 x 85.5 mm, thickness approx. 1 mm
• Superhydrophobic glass surface
• Covered with a 0.06 mm (60 micron) high spacer. This spacer has 96 recesses of 5 mm diameter. They are consistent with the SBS format for robotic handling. The spacer has an adhesive surface to which the hydrophobic coverslip attaches. The crystallization drops are, hence, sealed in these reaction chambers.

Description

Crystal Clear Sealing Film is an optically transparent sealing film for SBS format 24, 48, 96 and 384 well plates. The 2 mil +/-10% (0.05 mm) thick, optically clear, non-fluorescing polyester film with 1.0 – 1.3 mil (0.025 mm) thick acrylic custom adhesive is designed for the convenient and secure sealing of crystallization plates. A split, optically opaque 2.0 mil +/-10% polyester backing with two end tabs assures a clean uniform adhesive layer and allows for easy, accurate positioning on the plate. Before use, peel off the opaque center protective polyester backing to reveal the optically clear sealing film. Film length: 5.625″ +/- 0.015 Film width: 3.125″ +/- 0.015 End tabs: 0.375″ +/- 0.015 Perforations: 0.415″ +/- 0.015 from edge of film Corner radius: 3/32″. Single coated, 2 mil (0.05 mm), optically clear polyolefin film with pressure-sensitive silicone-based adhesive. Supplied as a 140 mm x 78 mm film with white polyester release liner (backing). Actual seal area after end tabs removed is 116 x 78 mm Seals well to polypropylene, polystyrene, and polycarbonate plates. Compatible with aqueous and organic solvents. Suitable for use between -70 and 110°C. For best seal, the plate must be sealed with the Film Sealing Paddle HR4-513. ClearSeal Film has a limited shelf life. Must be used before expiration date printed on box.

Each pack of film (HR4-521) includes one free Film Sealing Paddle (available separately as HR4-413). HR4-413 Film Sealing Paddle, 3 7/8″ x 2 15/16″, polyethylene, white. 5 paddles per pack. An alternative to the plate roller or brayer for pressing films to assure a secure uniform seal around all wells on 96-well, 384-well, and 1536-well plates. These paddles are especially recommended for sealing films on raised-rim plates because they fit within the rim of the plate.

Store ClearSeal Film at 20°C +/- 10 degrees at 50% RH +/- 10%.

Helpful Tips

  • Sufficient pressure must be applied to activate the silicone adhesive
  • Multiple swipes of the sealing paddle in both directions is highly recommended
  • A 10 minute or greater wait period before using for thermal cycling increases the film-to-plate bond

Description

A 38 µm soft non-permeable aluminum foil sealing film with strong medical-grade adhesive, AlumaSeal II sealing films eliminate the need for heat-sealing devices or mats during the resealing of reagents in polypropylene deep well blocks.

Each sealing film measures 82.6 x 142.9 mm and offers sufficient sealing area for 96 deep well blocks. Length between the perforations with end tabs removed is 125.4 mm. Compared to other aluminum foils, AlumaSeal II has less tendency to roll back on itself when removing the backing paper and it conforms well to the plate during application.

AlumaSeal II is a soft, pierceable adhesive film designed for the convenient and rapid sealing of polypropylene deep well blocks. A multiple split backing with two end tabs allows for easy, accurate positioning and secure sealing. Description

Siliconized glass cover slides allow a droplet to be suspended in a position which provides near optimal conditions for vapor diffusion with the surrounding reservoir solution.

These siliconized glass cover slides provide a consistent, high quality finish for crystallization experiments. The hydrophobic surface produces a drop which “stands well” and does not flatten on the glass. The siliconized surface prevents the adhesion of crystals and precipitate onto the glass surface. Use Dow Corning Vacuum Grease (HR3-510) or Dow Corning 7 Release Compound (HR3-508) to seal cover slide to plate. Available in 12, 18 or 22 mm diameter circles and 22 mm diameter squares. Available in 0.22 mm or 0.96 mm glass thickness. The 0.96 mm thick slides are virtually unbreakable during crystallization handling.

Description

Siliconized glass cover slides allow a droplet to be suspended in a position which provides near optimal conditions for vapor diffusion with the surrounding reservoir solution.

These siliconized glass cover slides provide a consistent, high quality finish for crystallization experiments. The hydrophobic surface produces a drop which “stands well” and does not flatten on the glass. The siliconized surface prevents the adhesion of crystals and precipitate onto the glass surface. Use Dow Corning Vacuum Grease (HR3-510) or Dow Corning 7 Release Compound (HR3-508) to seal cover slide to plate. Available in 12, 18 or 22 mm diameter circles and 22 mm diameter squares. Available in 0.22 mm or 0.96 mm glass thickness. The 0.96 mm thick slides are virtually unbreakable during crystallization handling.

Description

The Cover Slide Gizmo dispenses siliconized or plain slides for convenient drop building during hanging drop vapor diffusion crystallization. Load the cover slides into the dispenser. A clear plastic cover or sealed dispenser keeps slides clean. Slide the loaded dispenser along the platform and six cover slides are dispensed onto the platform. A black background on the platform makes for easy drop visualization as one builds the drop(s). Use finger or forceps to place the completed cover slides onto the crystallization plate.

Each Cover Slide Gizmo includes a single cover. Manual operation, no power required. See the Glass Cover Slide Gizmo Dispenser at work at minute 1:06.

Description

RNase-free, hydrophobic covers are lighter weight than glass. Polycarbonate plastic cover slides are ready to use without washing or siliconization. Hydrophobic working surfaces are protected with clean release liners to prevent RNase contamination. The plastic cover slides remain flat and will not curl, even at high temperatures. Plastic cover slides do not chip, crack, or break and are dust-free. Square cover slides are supplied protected in a sandwich polyethylene film. The circle cover slides are packed between fiber-free paper slips. Use Dow Corning® Vacuum Grease (HR3-510), Dow Corning® 7 Release Compound (HR3-508), or immersion oil to seal cover slide to plate.
