Accucore Vanquish UHPLC columns

Accucore Vanquish next-generation UHPLC columns from Thermo Scientific have 1.5um solid core particles, which combine the benefits of a solid core material with the increased chromatographic efficiency of a sub 2um particle. The high efficiency of Accucore Vanquish UHPLC columns enables the resolution of very complex mixtures. The columns are particularly effective when used with the Thermo Vanquish UHPLC system. A C18 phase is currently available with dimensions 100 x 2.1mm.

TLC Channel Plates

Macherey-Nagel now offer TLC Channel plates (20 x 20cm) with 19 channels, which help to prevent cross contamination when separating several samples. More samples can be separated on a plate and spot areas can more easily be determined. In addition, channel plates provide a kieselguhr zone for rapid sample application. Since kieselguhr is completely inert towards a large number of compounds, the samples always form a narrow band at the interface of the two adsorbents, irrespective of shape, size or position of the spots in the concentrating zone. Separation then takes place in the silica layer. This product was developed to be a direct alternative to the discontinued Whatman TLC channel plates.

Biomolecule Purifications Using CIM Monoliths CIM (convective interaction media) monolithic columns are an innovative chromatographic technology that can redefine biomolecule purifications at all scales from laboratory to industrial GMP manufacture. Suitable application areas include virus production/purification, purification of plasmids and proteins including His-tagged recombinant proteins and antibodies such as IgG and IgM. CIM monolithic columns operate at up to 10 times higher flow rates than comparable particle based supports, without comprising resolution or dynamic binding capacity. The media offers high dynamic binding capacities for large biomolecules, which it can accommodate due to the relatively large pore size of the interconnected network of flow-through channels. These channels can be functionalised with IEX, HIC, affinity or customised ligands. TOYOPEARL NH2-750F Resin

TOYOPEARL NH2-750F Resin Tosoh Bioscience have introduced a new salt tolerant ion-exchange resin – TOYOPEARL NH2-750F. This resin provides high protein binding capacity at elevated ion strength and shows unique selectivity, differing from conventional anion-exchange material. It features larger pore size than Tosoh Bioscience’s standard TOYOPEARL 650 resins, providing access for larger molecules such as aggregates. The smaller particle size (45μm) is similar to the TOYOPEARL Protein A resins.

Centre Draining Vials & Bottles from MicroSolv

Centre Draining Vials & Bottles from MicroSolv MicroSolv have introduced a new product line of Centre Draining Maximum Recovery (CDMR) vials and bottles. These are designed with screw caps and have a conical bottom to allow the contents to drain completely. They are made from Type 1 borosilicate glass and are useful in library storage, extraction, concentration, small scale reactions and liquid-liquid extraction.

HALO-2 columns for UHPLC

HALO-2 columns for UHPLC The new HALO-2 columns from Advanced Materials Technology are based on their Fused-Core technology, but with a smaller particle size of 2m. These columns offer the advantages of sub 2m fully porous particles but at lower operating pressures. Manufactured with 1m column inlet frits, HALO-2 columns are less susceptible to column plugging than typical 0.2-0.5m frits on sub 2m fully porous columns. HALO-2 columns can be used up to 1000 bar but will actually produce 20% lower back pressure than most commercially available sub 2m UHPLC columns under the same conditions. There are two phases currently available: C18 and PFP. A 4-page flyer and three application notes with HALO-2 C18 are also available.

New Chromolith phases

New Chromolith phases Merck Millipore have expanded their range of Chromolith phases to include Phenyl, DIOL and CN. As for the earlier C18, C8, Si and NH2 bonded Chromolith phases, these newer columns are particularly advantageous for the analysis of ‘dirty’ samples. Chromolith DIOL is useful as an intermediate polarity phase between C18 and Silica and can be used with both NP and RP solvents.

SOLAu SPE Plates

SOLAu SPE Plates Thermo Scientific’s SOLA is the first fritless SPE product range. The proprietary manufacturing process involved in the production of SOLA eliminates issues that can be associated with conventional loose-packed SPE products such as settling and voiding in production or transportation, which can create phase channelling and packing irreproducibility. SOLA fritless SPE products combine the polyethylene frit material and the media components into a uniform sorbent bed, thus removing the need for frits. Thermo Scientific have now expanded their fritless SPE product range to include SOLAu plates. These are designed for bioanalytical and clinical research analysts who are working with reduced sample volume analysis or require pre-analysis sample concentration.

AVCS Caps from Thermo Scientific

Thermo Scientific have introduced AVCS caps which provide greater sealing capacity than conventional screw caps, so that evaporation is minimised and septa push through is prevented. These are a cost effective alternative to the use of caps with bonded septa.

NUCLEOSHELL RP 18plus

NUCLEOSHELL RP 18plus is a new core-shell C18 phase from Macherey-Nagel. It is available with particle sizes of 2.7 and 5m. Due to its monomeric bonding this phase offers hydrophobic characteristics with distinct polar selectivity. A special derivatisation process generates a medium density bonded silane with reduced steric selectivity compared to NUCLEOSHELL RP 18. Whereas NUCLEOSHELL RP 18 has an operable pH range of 1-11, the recommended pH range for NUCLEOSHELL RP 18plus is 2-9. NUCLEOSHELL RP 18plus is recommended for polar applications eg. antibiotics, water-soluble vitamins and organic acids.

Nacalai Tesque have recently introduced COSMOCORE C18, a 2.6um core-shell phase with a solid core of 1.6um and a shell of 0.5um. COSMOCORE 2.6C18 enables ultra high performance LC analyses with conventional HPLC equipment. Columns can produce the same efficiency as sub-2um columns, but operate at less than half the pressure. They show excellent stability over the pH range of 1.5 to 10. Nacalai Tesque claim that COSMOCORE C18 features high retention compared to other core-shell columns. In addition, the phase exhibits high loading capacity, a benefit when analysing trace components. Columns with i.d.s of 2.1, 3.0 and 4.6mm are currently available. Chiral Technologies are about to launch their first medium pressure liquid chromatography columns (MPLC). These new CHIRALFLASH columns are packed with immobilised chiral stationary phases for the separation of enantiomers. The columns, with a length of 100mm and i.d. of 30mm, are packed with Daicel’s 20um CHIRALPAK IA, IC, ID and IF materials. They are specifically designed for rapid and low cost separation of target compounds for exploratory stage processes. The columns are reusable and compatible with any commercially available flash chromatography system.

MPLC method development is first carried out on analytical CHIRALPAK HPLC columns (20um, 100 x 4.6mm) before scaling up to CHIRALFLASH MPLC columns. A 6-page product brochure is available.

YMC have launched new Meteoric Core core-shell columns for ultra fast separations with outstanding resolution. YMC Meteoric Core columns are initially available in three phases, 80Å trifunctionally bonded C18 and C8 phases and a 160Å trifunctionally bonded C18 BIO phase for protein and peptide analysis. ACE UltraCore is a new range of ultra-inert solid-core (superficially porous) particle UHPLC/HPLC columns from Advanced Chromatography Technologies Limited.

The monodisperse particle distribution of these phases combines high efficiency and performance, with a low column back pressure, to deliver exceptional chromatography. ACE UltraCore is available with SuperC18 and SuperPhenylHexyl bonding. Both phases feature proprietary Encapsulated Bonding Technology (EBT), which results in excellent peak shape and superb phase stability across an extended pH range of 1.5 to 11.0. Encapsulated Bonding Technology dramatically increases the ligand coverage of the silica surface and effectively eliminates the effect of unbonded silanol groups on separations. This higher ligand coverage results in improved inertness, chromatographic performance and stability.

COSMOSIL PFP and PBr HPLC columns

Nacalai Tesque have launched COSMOSIL 5PFP, a pentafluorophenyl-bonded HPLC column with alternative selectivity to C18 columns. In addition to a hydrophobic separation mechanism, the phase offers dipole-dipole and p-p interactions. COSMOSIL 5PFP shows good retention for compounds with strong dipole moments, such as cationic or halogenated compounds, and exhibits high steric selectivity for structural isomer separations. Columns with analytical to preparative dimensions are available.

Cogent Diol

is the latest addition to the MicroSolv TYPE-C Silica™ range of stationary phases. This 4mm diol bonded phase is suitable for the reversed-phase or aqueous normal-phase (ANP) analysis of polar compounds and is LC-MS compatible.

Columns show fast equilibration between gradient runs and can be successfully used in studies of pathways in human pathology.

Cogent UDA

is a new unique selectivity phase based on MicroSolv’s TYPE-C Silica and silica hydride surface chemistry. This silica hydride surface is bonded with an eleven carbon chain terminating in a carboxylic acid (undecanoic acid), giving the phase weak cation-exchange properties in addition to aqueous normal-phase (ANP). All the features and benefits of TYPE-C Silica, such as fast equilibration, precision, multi-mode and robust column-lifetime are maintained.

The Cogent UDA columns have applications in metabolomics and have been used for the analysis of nucleotides.

For decades, liquid chromatography stationary phase design has been dominated by the idea of elimination of multiple, or “unwanted”, interactions that occur in mixed-mode separations. Reverse-phase chromatography is making the point of eliminating silanol interactions with amine-containing analytes by developing base-deactivated phases.

Within the same trend, adsorption interactions are considered undesirable as opposed to steric interactions in size-exclusion chromatography, and non-ionic interactions are generally viewed as complications in ion-exchange and ion-exclusion chromatography.

However, there are ways to benefit from multiple interactions on the stationary phase. SiELC introduces Primesep™ HPLC columns, specifically designed for mixed-mode separations and capable of separating a tremendous range of compounds by different separation modes based only upon mobile phase selection.

The improved selectivity for peptides on the Vydac MS columns results in better primary structure definition and easier identification of degradation products and other protein characteristics.
Vydac 218MS and 238MS are used for simple enzymatic digests (< 12 proteins) or biomolecules (0-5kDa).

Vydac 208MS is ideal for biomolecules of molecular weight 5-10kDa.

Vydac 214MS has lower capacity than C18 or C8 and is suitable for hydrophobic proteins or when minimal organic solvent is required. It is recommended for biomolecules of > 10kDa molecular weight, undenatured intact proteins, antibodies, oligonucleotides, human growth hormone.

Vydac Everest

Everest columns have unique selectivity and sensitivity, which are the result of bonding technology that improves C18 surface coverage and deactivates residual silanols. Previously, the best 300A C18 chemistries have had carbon coverage in the 2.8 to 3.6umol/m2 range. Everest C18 coverage is in excess of 4umol/m2 and approximates the theoretical limit based on surface area. The increased shielding of the base silica increases column lifetime and reduces the amount of TFA required to shield the silica.

Wakopak Fluofix Phases

Wakopak Fluofix phases are bonded with branched chain perfluorohexyl groups on 120Å pore size silica, in both endcapped and non-endcapped options. They exhibit increased retention and selectivity compared with C18 phases for compounds containing fluorine and chlorine substituents. This is due to the carbon-fluorine bond being more polar than the carbon-hydrogen bond of C18 phases.

Wakopak Fluofix phases are also highly selective for the analysis of non-halogenated polar compounds containing hydroxyl, carboxyl, nitro and other polar groups. This is most apparent when the functional groups are located on an aromatic or other rigid ring system.

NUCLEODEX®
NUCLEODEX® phases are manufactured by chemically bonding one of three cyclodextrins to NUCLEOSIL 100 silica. Cyclodextrins are cyclic oligosaccharides. The size of the chiral ring and interaction of racemic mixtures with the chiral sugar units control the enantiomeric separation process. In the permethylated cyclodextrins all hydroxy groups are replaced by methoxy groups. Alternative selectivities are obtained. NUCLEODEX® screening kits for rapid and economical method development are available. Each kit contains a 30 x 4mm cartridge of each of the four NUCLEODEX materials, β-OH, α-PM, β-PM and γ-PM, together with a holder.

RESOLVOSIL®
RESOLVOSIL® BSA-7 was one of the first commercially available protein-bound columns to demonstrate optical resolution.


NUCLEOSIL® chiral-1
NUCLEOSIL® chiral-1 is a unique material offering separation of racemic molecules by ligand exchange HPLC. An L-hydroxyproline Cu2+ complex is chemically bonded to NUCLEOSIL 120 silica. Molecules containing two correctly spaced polar functional groups, such as α-amino acids can be optically resolved by this column through the formation of diastereomeric complexes of differing stability.

MACHEREY-NAGEL have introduced the NUCLEOCEL series of polysaccharide chiral stationary phases. The chiral selector of NUCLEOCEL DELTA is cellulose-tris-3,5-(dimethylphenyl) carbamate and together with the supramolecular helical polysaccharide surface structure is mainly responsible for the chiral recognition mechanism. For NUCLEOCEL ALPHA the amylose counterpart of tris-3,5-(dimethylphenyl) carbamate performs this task.

The NUCLEOCEL product line includes columns for normal-phase and reversed-phase applications. The NUCLEOCEL DELTA and ALPHA phases are intended for analyses using normal-phase eluents, typically heptane – IPA. The NUCLEOCEL DELTA-RP and ALPHA-RP phases are designed for use under reversed-phase conditions either in polar organic mode or with eluents containing high concentrations of chaotropic salts such as perchlorate.

In addition to the 10μm phases (DELTA and DELTA-RP), higher resolution 5μm phases are available for these and for the ALPHA and ALPHA-RP phases.

NUCLEOCEL chiral stationary phases offer high loading capacities and thus are suitable for scale-up methods. The stable silica support protects the coated polysaccharide and tolerates high flow rates with a maximum operating pressure up to 2000 psi.

NUCLEODUR® C18 Isis

NUCLEODUR C18 Isis is a C18 bonded phase with specially cross-linked surface modification. This polymeric bonding leads to high steric selectivity and is therefore suited for separation of positional and structural isomers and planar/non-planar molecules. The phase has an enhanced pH stability of 1 to 10. Figure below demonstrates the increased steric selectivity of NUCLEODUR C18 Isis for o-terphenyl (non-planar) and triphenylene (planar) compared with a monomerically bonded C18 phase.

NUCLEODUR® C18 Pyramid

NUCLEODUR® C18 Pyramid is a C18 bonded phase with hydrophilic endcapping, designed especially for use with eluents containing up to 100% water. The phase also shows improved retention characteristics for very polar molecules, such as short chain organic acids.

NUCLEODUR® Sphinx RP is a bifunctional phase consisting of a well-balanced ratio of octadecyl and phenyl groups covalently bonded to the NUCLEODUR silica. The combination of classical hydrophobic with π-π interactions expands the scope of selectivity in comparison with conventional reversed-phase materials. NUCLEODUR Sphinx RP is particularly suited for the separation of molecules containing aromatic and multiple bonds. It may also outperform standard C18 phases for the separation of polar compounds. Figure below shows a comparison of the separation of flavonoids on three different NUCLEODUR phases.

NUCLEODUR® HILIC

NUCLEODUR HILIC is a zwitterionic modified stationary phase. The betaine character of the ammonium – sulphonic acid ligands results in an overall neutrally charged, but highly polar surface. This column is ideal for analysing polar analytes such as acids and bases, natural compounds, nucleosides, amino acids, water soluble vitamins etc. Due to its high loadability, NUCLEODUR HILIC is suitable for preparative applications.

An increase in plate number of approximately 67% in transferring from 3μm to 1.8μm particles offers the possibility of using shorter columns with equal plate numbers, resulting in a decrease in analysis time. Typically, for a 50 x 4.6mm NUCLEODUR C18 Gravity column:

3μm: N ≥ 100,000 plates/m

1.8μm: N ≥ 166,667 plates/m

Improvement in resolution

The use of 1.8μm instead of 3μm particles can lead to an increase in resolution of approximately 30%. This is illustrated in Figure below for the separation of naphthalene and ethylbenzene.

All of the NUCLEODUR® premium phases are now available in 1.8μm particle sizes. Columns packed with these microspherical particles show improvements in terms of plate numbers, column efficiencies and resolution, compared with their 3μm counterparts.

Column back pressure

The smaller particle size results in an increased back pressure. However, the high sphericity of the NUCLEODUR particles and their very narrow particle size distribution means that back pressure is kept at a moderate level. In addition, the particles are fractionated in order to limit the increase in back pressure. Nevertheless, the use of columns packed with sub 2μm particles generally makes special demands on the HPLC equipment. Pumps should be designed for pressures of 250 – 1000 bar and the entire system should feature the lowest possible dead volume, for optimum column performance.

Higher flow rates and shorter run times

The optimal flow rate for 1.8μm particles is higher than for 3 and 5μm particles.

NUCLEODUR® is an ideal material for scaling-up methods due to the availability of a wide range of particle sizes. The material provides a high mechanical bed stability and the narrow particle size distributions provide lower pressure drop due to the absence of fines. The material is reusable even after several cycles of repeated packing.
